Although unable to be elucidated analytically, theory suggests preferred orientations of cis-12-OH-TBOH (relative to C13 methyl) and trans-5-OH-TBOH, with the former most thermodynamically stable overall. Intravaginal sponges impregnated with progesterone and synthetic progestins may have some degree of success in synchronizing estrus in mares.16 Unfortunately, sponges have the potential to adhere to the vaginal wall and induce necrotic vaginitis. Intravaginal devices that release progesterone slowly have been investigated in Europe and Australia and hold promise for future use. Microspheres containing progesterone and progesterone plus estradiol-17β have been injected to control or synchronize the estrous cycles of mares. Some long-acting preparations of progesterone plus estradiol were available by prescription from a pharmacy in Kentucky but the FDA recently seized compounded equine drugs. The FDA understood the pharmacy was illegally manufacturing drug products from bulk ingredients without FDA approval and selling them outside a valid veterinarian-client-patient relationship. The natural sex steroid hormones estradiol, testosterone, and progesterone may be in use illegally for growth promotion.
Studies on the metabolism of 17β-estradiol in bovine species indicate the formation of lipoidal esters, disposed particularly in body fat. These lipoidal esters show a high oral bioavailability in rodent experiments; thus, the consequence of their consumption needs to be considered in a risk assessment (RA). It is known that experiments with heifers indicated a dose-dependent increase in residue levels of all hormones particularly at the implantation sites. Misplaced implants and repeated implanting, which seem to occur frequently, represent a considerable risk that highly contaminated meat could enter the food chain (EC, 2002a). Hancock and colleagues18 reported that infused E2 has a short half-life of 7.7 minutes in the fast pool and a longer half-life of 41.5 minutes in the slow pool. Little is known, however, of the effects of the synthetic steroids on wool growth. Coehlo, Galbraith & Topps (1981) reported a reduction in wool per unit body weight in sheep implanted with trenbolone acetate and oestradiol-17β, suggesting lowered efficiency of wool growth. Similarly Yasin & Galbraith (1981) noted that trenbolone acetate, in combination with either oestradiol-17β or zeranol, reduced wool output per unit body weight. Neither of these trials attempted to isolate the effects of trenbolone acetate alone from those of the androgenplus-oestrogen combination. Use of TBA and EB combination implants, in this study, resulted in increased anabolism as suggested by the observed reduction in serum concentration of urea-N and increased serum concentration of IGF-I compared with NI steers. These results indicate that the use of a lower dose implant containing 100 mg TBA + 14 mg EB can result in comparable growth performance to an implant containing 200 mg TBA + 28 mg EB.
